Extraction of extracellular polymeric substances from dam lake fresh sediments derived from crystalline bedrock

چکیده

Extracellular polymeric substances (EPS) produced by microorganisms have a key role in the sedimentary compartment, e.g. promoting aggregation and biostabilisation of sediment particles increasing chemical reactivity at water/sediment interface. Therefore, proper extraction methods are needed to study this EPS matrix. In work, nine based on physical (centrifugation, sonication), (sodium hydroxide, sodium pyrophosphate, tetraborate), both (cation exchange resins, i.e. CER) treatments their combinations, as well solid:liquid ratio used for extraction, were compared quantity compositions extracted EPS. The organic carbon was quantified nature biochemical macromolecules (proteins, polysaccharides, humic-like compounds) evaluated using colorimetric methods. amount ATP an indicator cell lysis showed contamination with intracellular materials Moreover, presented large impurities due non-removal reactant salts ultracentrifugation. Extraction of Structural Extracellular Polymeric Substances from Aerobic Granular Sludge

To evaluate and develop methodologies for the extraction of gel-forming extracellular polymeric substances (EPS), EPS from aerobic granular sludge (AGS) was extracted using six different methods (centrifugation, sonication, ethylenediaminetetraacetic acid (EDTA), formamide with sodium hydroxide (NaOH), formaldehyde with NaOH and sodium carbonate (Na2CO3) with heat and constant mixing). AGS was ...

Different Types of Diatom-Derived Extracellular Polymeric Substances Drive Changes in Heterotrophic Bacterial Communities from Intertidal Sediments

Intertidal areas support extensive diatom-rich biofilms. Such microphytobenthic (MPB) diatoms exude large quantities of extracellular polymeric substances (EPS) comprising polysaccharides, glycoproteins and other biopolymers, which represent a substantial carbon pool. However, degradation rates of different EPS components, and how they shape heterotrophic communities in sediments, are not well ...

Characterization of extracellular polymeric substances from acidophilic microbial biofilms.

We examined the chemical composition of extracellular polymeric substances (EPS) extracted from two natural microbial pellicle biofilms growing on acid mine drainage (AMD) solutions. The EPS obtained from a mid-developmental-stage biofilm (DS1) and a mature biofilm (DS2) were qualitatively and quantitatively compared. More than twice as much EPS was derived from DS2 as from DS1 (approximately 3...

Redox properties of extracellular polymeric substances (EPS) from electroactive bacteria

Although the capacity for electroactive bacteria to convert environmental metallic minerals and organic pollutants is well known, the role of the redox properties of microbial extracellular polymeric substances (EPS) in this process is poorly understood. In this work, the redox properties of EPS from two widely present electroactive bacterial strains (Shewanella oneidensis and Pseudomonas putid...

Bedload-to-suspended load ratio and rapid bedrock incision from Himalayan landslide-dam lake record

About 5400 cal yr BP, a large landslide formed a N400-m-tall dam in the upper Marsyandi River, central Nepal. The resulting lacustrine and deltaic deposits stretched N7 km upstream, reaching a thickness of 120 m. C dating of 7 wood fragments reveals that the aggradation and subsequent incision occurred remarkably quickly (∼500 yr). Reconstructed volumes of lacustrine (∼0.16 km) and deltaic (∼0....
